At around 6.40 am, a level crossing at Barwon Terrace in South Geelong witnessed a collision between a truck and a Melbourne-bound V/Line train.
The truck driver had serious injuries that required immediate medical attention, but he passed away at the spot.
There were no passengers on board, and the driver was unharmed.
All trains on the Geelong line are suspended due to the incident, which has caused significant delays for commuters.
Between Waurn Ponds and Geelong, buses now run in place of trains.
Authorities are asking for any witnesses or anyone with dashcam footage to come forward as they are still investigating the collision.
